Production, Optimization and Purification of Laccase Produced by Pleurotus ostreatus MH591763

Abstract


The optimization of production medium for laccase produced by Pleurotus ostreatus (Gene Bank Accession number MH591763) to reduce the cost of laccase production in large scale is the basic research in industrial field. In this study, the laccase culture medium was optimized for growth parameters such as incubation period, pH, temperature, carbon and nitrogen source on fungal growth and enzyme production. Purification and characterization of the enzyme was done by following methods: Acetone precipitation, Dialysis, Column Chromatography and SDS PAGE. The maximal laccase activity 543±19 U/l was observed on 10th day of incubation, pH shows 538±1.1 U/l at 6.0, temperature shows 516±2.5 U/l at 40°C. The laccase activity on maltose 519±1.1 U/l act as best carbon source and yeast extract 592±1.5 U/l was found as the excellent nitrogen source for high amount of laccase expression by P. ostreatus. In SDS PAGE Molecular weight of laccase 65 kDa was confirmed with standard molecular markers.
