Abstract

Based on the global fame of benzimidic acid/benzoic acid derivatives in exhibiting antimicrobial activities against a variety of pathogens (particularly nosocomial infections), two novel compounds (S1 and S2) of benzimidic acid scaffold were rationally designed from a hydrazine derivative and fabricated using the conventional synthesis technique. Through sophisticated instruments, the molecules were comprehensively characterized and investigated for antibacterial activity against pathogenic Escherichia coli strain (MTCC 2961). The novel benzimidic compound S1expressed relatively low bactericidal activity against E. coli at all three concentrations that is 20 μg/ml (11 mm), 40 μg/ml (12 mm) and 60 μg/ml (14 mm) as compared to the standard marketed drug ciprofloxacin which displayed zone of inhibition (ZOI) of 32.1 mm at 10 μg/ml concentration. In contrast to this, the compound S2 expressed drastically high bactericidal activity against pathogenic E. coli species with at all three concentrations that is 20 μg/ml (38 mm), 40 μg/ml (41 mm), and 60 μg/ml (46 mm). The meta-substituted compound exhibited higher anti-microbial activity as compared to the standard marketed product ciprofloxacin. Although the compounds were rationally designed and synthesized after lots of literature survey and computational aid, but the results came serendipitously where the para-substituted analog failed completely but the meta-substituted analog demonstrated remarkable activity.
