Production of High Yield Amylase and Protease by Aspergillus oryzae using Solid-State Fermentation

Aspergillus oryzae strains were isolated from different sources and its ability to produce industrial scale amylase and protease using solid-state fermentation was carried in the present study. Different fermentation substrates such as rice bran and wheat bran were evaluated for maximum enzyme yield. Enzyme production was maximum on rice bran and it was achieved with optimized fermentation parameter such as temperature 30°C, pH 4–6, moisture content 60%, inoculum size 0.2 mL (24 million spore/gram) and 48 h incubation time. Under these conditions, amylase activity reached to maximum level of 23,769 U/g and protease activity 2,063 U/g. The quantity of enzyme achieved using this strain is highly significant compared to already reported yiled of these enzymes using different substrates. Hence, the present study is successful in isolating Aspergillus spp. having high yield industrial enzyme production in standardized fermentation parameters.
