Improvised Rapid Antibiotic Sensitivity Test by Disc Dispersion Method

Multidrug resistance is the one of vital issue globally, there are several causes for multidrug resistance, like improper utilization of antibiotics and selection of suitable antibiotics are the one of the predominant reasons for multidrug resistance gaining. To enable physician to select the suitable antibiotics, appropriate antibiotic sensitivity shall be performed in shortest time period. A method has been developed to get the antibiotic sensitivity test in 2–3 hours for Escherichia coli bacteria. In this method culture media has been modified to get the faster results and helps the organisms to be there in log period for extended time. This method gives quick results and it is very easy to read the results by color change, less sophistication and less skill required for results interpretation. This is called as “improvised rapid antibiotic sensitivity test by disc dispersion method”. This has been tested on the Escherichia coli bacteria, total 99 clinical isolates were collected from pathological testing laboratories in Hyderabad city and antibiotic sensitivity profile determined by both conventional “Kirby and Bauer Method” and “Improvised Rapid Antibiotic Sensitivity Test by Disc Dispersion Method”. All challenged isolates demonstrated comparable results, 92.8% of E. coli isolates tested were shown similar results. By using statistical tool “Chi-Square Tests”, p Value, Sensitivity (%), Specificity (%) and Accuracy (%) was calculated. Two methods demonstrated similar results like sensitive as sensitive and resistance as resistance with comparable accuracy. Level of significance was considered for p value as 0.05 and it was observed that the p Value was >0.05. >80% was considered as significance for Specificity, Selectivity and Accuracy.it was observed that obtained % for selected parameters were >80% and this indicates the both methods are suitable and comparable. The basic principle of improvised method is similar to the macro broth dilution method but it is completely different to the existing method in terms of experimental design, process and interpretation. This improvised antibiotic sensitivity test method will enable the technician to obtain results with minimum efforts. By observing color change in the culture media and turbidity, one can determine antibiotic sensitivity profile of the microorganism (sensitive or resistant). This method is comparatively economic and less sophisticated.
