Evaluation of Cholesterol Reduction in Probiotic Strain Lactobacillus brevis isolated from Human Fecal Sample by Using Surface Response Methodology

Lactobacillus brevis, isolated from human feces was assessed for its in vitro cholesterol reduction ability. The effects of bile salts, inoculum size and contact time in simulated intestinal juice were studied. Central composite design model was utilised to determine the effects of interactions of significant variables on cholesterol reduction. By developing a linear regression model, as a function of growth associated variables in vitro cholesterol reduction was assessed. The results demonstrated that bile salts had significant effect in cholesterol reduction and maximum reduction, 58% was obtained at the central point. Using RSM, a predictive model was developed to ascertain the cholesterol reduction. R2 (0.9588) close to 1.0, indicates that the model is strong. F value of 25.84 and values of probability > F (<0.0001) indicate that model terms are significant. The developed and validated regression model can be efficiently applied for assessment of cholesterol reduction by L. brevis.
